Morphological indicators and those of reversible aggregation of red blood cells were studied in postnatal development of white mice. The linear correlation coefficient made it possible to estimate the interrelationship between the indicators of peripheral blood and marrow in mice, to describe the age dynamics of blood system indicators, and to determine the most informative indicators in the process of postnatal development of white mice.

It was established that quinoid products of tyrosine oxidation, analogues of natural radiotoxins, when used in the studied doses decreased the radiosensitivity of spleen stem cells and enhanced the restoration of bone marrow cellularity of mice exposed to sublethal gamma-radiation doses.

A study was made of the number of CFUs and karyocytes in thigh bone and the concentration of functional cells in the peripheral blood of mice subjected to fast neutron- and gamma-irradiation four times at a 60-day interval (210 cGy per fraction). The regenerating potential decreased and the half-recovery time T1/2 increased in the haemopoietic tissue as the number of fractions and total absorbed dose increased. The dependence of T1/2 on the equivalent radiation dose was as follows: T1/2 = T1/2(0)e0.0009D.

It has been revealed by competitive repopulation assay that hemopoietic stem cells capable of supporting long-term hemopoiesis in the culture failed to regenerate after irradiation. 19 weeks after irradiation with 4 Gy the content of hemopoietic stem cells was 0.5% normal, while regeneration of CFUs was achieved up to subnormal level.

1747. [Pentagastrin as a stimulator of immunogenesis].

It has been shown that subcutaneous injection of pentagastrin to mice in doses 0.01-5 micrograms per animal during 10 days resulted in a considerable stimulation of the immune response to sheep red blood cells (SRBC). Pentagastrin in doses of 5 and 1 micrograms per animal was demonstrated to have the highest immunostimulating effect. These doses increased the production of IGM-antibody-forming cells 2.2-2.7-fold and produced a twofold elevation of the antibody titer. Pentagastrin did not influence the immune response to thymus independent Vi-antigen. The in vitro treatment of mouse bone marrow cells with pentagastrin (0.1 or 0.01 microgram/ml) increased the number of Thy-1 positive cells from 0 to 16-17%. Pentagastrin at a dose of 0.001 microgram/ml was not effective.

Proliferative activity of hemopoietic bone-marrow stem cells has been studied in splenectomized mice in response to sheep red blood cells (2 X 10(8], pneumococcal polysaccharide (100 micrograms) and lipopolysaccharide E. coli (100 micrograms) injections. Spleen and its lymphoid cellular elements were shown to be of great importance for the regulation of the functional activity of hemopoietic stem cells under the antigenic influence.

The influence of the duration of the organ cultivation on the protective potential of mouse fetal liver hemopoietic cells was investigated. The protective potential was evaluated according to a 3-week viability of the lethally irradiated (mean LD88/21) mice following syngeneic transplantation. During 20 days of cultivation the protective potential (calculated per number of injected cells) remained at the initial level. With more prolonged cultivation (24-62 days) the protective potential was retained only in part of cultures and the mean effect was reduced. No parallelism has been revealed between the totipotent hemopoietic precursors capable of replacing the hemopoietic cells of an irradiated recipient and the CFUs in long-cultivated fetal liver cultures.

The exogenous administration of cAMP to nonirradiated mice slightly stimulates the haemopoietic stem cell migration, does not promote colony-formation in the spleen, in conditions of the endotest, and has no therapeutic effect on the irradiated rats and hamsters.

Acetylcholine and low concentrations of nicotine were shown to increase firing rate, to induce grouping of spikes of spontaneously active cells and to activate "silent" neurons. High concentrations of nicotine exert a biphasic (excitatory-inhibitory) effect. Atropine just alters and benzohexonium suppresses the spontaneous unit activity. Changes in the unit activity seem to stem from an interaction of cholinergic substances with the nicotine and muscarine cholinoreceptors. Neurons with a mixed type activity were found to receive inhibitory cholinergic projections.

A higher death rate of BALB/c as compared to CBA mice was registered during long-term external irradiation. The changes in the content of the morphologically recognized bone marrow cells and blood formed elements in the exposed mice of the studied lines were different.

A study was made of the yield of reciprocal translocations in stem spermatogonia of mice exposed to alpha-radiation (238Pu) and whole-body acute and chronic gamma-radiation within a wide range of doses and dose-rates. The frequency of reciprocal translocations induced by a single intraperitoneal administration of plutonium nitrate was relatively low and independent of the dose 1.5-18 months after the effect. The yield of the reciprocal translocations induced by chronic effect of gamma-radiation was appreciably lower than that observed after acute irradiation with the same dose and grew linearly with dose. The RBE of alpha-radiation was 10-20 with respect to chronic effect of gamma-radiation.

The data are presented on the stimulatory effect of interleukin-2 on the formation of spleen exocolonies from bone marrow irradiated in vitro with doses of 0.5 to 2.5 Gy. It is suggested that the effect observed is associated with the increased proliferation of CFUs survived after irradiation.
