The fraction of condensed chromatin (C. C., %) in cell nuclei images was determined by the aid of the "random step grid". 100 random points were thrown on every electron microscopic images. The number of points in C. C. images was counted. Lymphocyte nuclei in spleen and lymph nodes of normal cows have a higher content of C. C. (61 +/- 1.6%) Than those of cows suffering from lympholeucosis in haematological (49 +/- 21%) and in tumor (44 +/- 1.1%) stages. The counting procedure takes about 2 hours for 90 images. The random step grid can be easily available in every day practice.

4 types of marker chromosomes and of their combination in mouse ascite Ehrlich-I. Ch. Ph. tumor cells are described. Two most frequently encountered subpopulations of cells are found in the tumor: one--with A+B+2C and A+D+2D markers, and the other--with A1+A2+2B+D1+C markers. The former population dominated during 8 days, to be gradually substituted with the former subpopulation. Changes in the number of cells with certain chromosome number in the near-diploid and near-tetraploid cell zones were followed within one passage.
2149. [Comparative study of the karyotype and histostructure of neoplasms of the uterine body, stomach and malignant melanoma].
Direct analysis of karyotype revealed a heterogeneity of chromosome composition, variations in modal lines and chromosome aberrations, diversity of marker chromosome morphology. The variability of cytogenetic alterations was found in tumors both of the same and of different origin. These results established the nature of genome and chromosome alterations in the investigated tumors. The problem is discussed of a possible use of cytogenetic analysis for a more concrete characterization of human tumors in terms of their biological features.

The state of the major and the minor DNA grooves in purified mono- and oligonucleosomes and in the complexes of DNA with different histones have been studied by means of methylation of DNA with dimethyl sulphate. In nucleosomes histones shielded major groove by 18--20%. This result agrees well with our previous data obtained with chromatin, nuclei and whole cells. Each of the purified histones H2a, H2b, H3 and H4 as well as N-terminal peptides of H4 histone cause relative shielding of the DNA major groove by 15--18% like whole histone does. H1 histone protects neither DNA grooves from methylation. Our results suggest that histones are buried partly in the major groove of DNA in chromatin and purified nucleosomes. The arrangement of histones in the major groove does not depend probably on their specific organization in nucleosomes.
2155. [Complementary RNA in the chicken sarcoma cells and Rous sarcoma virus].
The subcellular localization in chicken Rous sarcoma of nucleotide sequence, complementary to Rous sarcoma virus RNA was examined by RNA/RNA molecular hybridization. The preparations of radioiodinated virion RNA were annealed with RNAs from different fractions (nuclei, mitochondria, free and membrane-bound polyribosomes) isolated from chicken Rous sarcoma. Formation of RNA-ase resistant hybrids between the viral 125I-RNA and RNA from the mitochondria and membrane-bound polyribosomes was revealed. The latter were characterized by a higher relative redundancy of nucleotide sequences complementary to virion RNA than that in the former, by factor 446. The role of complementary ribonucleotide sequences is discussed.
2156. [Expression of C-type oncornavirus proteins in tumors of CC57BR mice].
Murine endogenous oncornavirus (C-type) genome expression was investigated in methylcholanthrene-induced (MC-induced) tumours of low-leukemic CC57BR mice by means of the radioimmunodiffusion method, using the precipitating test-systems. The gs-1 antigen of murine C-type viral p30 protein and Gross leukemia virus type-specific antigen (AGLV) served as viral genome markers. The gs-1 antigen was found in the primary and continuous MC-induced sarcomas, whereas AGLV appeared only during the tumour passage. A distinct association was revealed between the quantity of p30 protein (gs-1 antigen titre) and the presence of AGLV. Simultaneously with the appearance of AGLV the gs-1 titre increased markedly and the AGLV disappearance was always accompanied by a decrease of the gs-1 titre. The data presented suggest a coordinated expression of the two investigated proteins of murine C-type endogenous oncornaviruses in the chemically-induced tumours of CC57BR mice.

A comparative karyologic analysis of three continuous cell lines of different origin (HEp-2, FL and RH) has shown them differing from each other both in the number of chromosomes, and in marker chromosomes. In cultures HEp-2, the modal class consists of cells possessing 58-59 chromosomes, cells in cultures Fl and RH have 60 and 66 chromosomes, resp. Marker chromosomes both similar and not similar, i.e. characteristic of a given cell line only were found in all the investigated cell lines. The data obtained do not exclude a possibility of contamination of studied continuous cell cultires with HeLa cell line.

The authors have determined the activity of N-acetyltransferase and the type of acetylation in patients with cancer of the lung (38) and gastrointestinal tract (39), and in 72 control individuals. It was found that there is a changed distribution of patients in relation to the activity of acetylation in maintaining the normal correlation of "slow" and "fast" acetylators. There was noted an increase in N-acetyltransferase activity with greater spread of the tumor process, whereas the activity decreased in radical operations and increased in palliative surgical procedures and in the presence of recurrences or metastases.
