921. [Comparison of the methods for assessing Ah effects in aquatic system by semipermeable membrane device and caged fish].
Semipermeable membrane devices (SPMDs) in combination with in vitro ethoxyresorufin-O-deethylase (EROD) assay was used to assess the aryl hydrocarbon receptor (AhR) effects in aquatic system. In present work, polycyclic aromatic hydrocarbons (PAHs) were selected as target compounds and SPMDs were deployed simultaneously with caged crucian carp for 32 days at five sites in Meiliang Bay, Taihu Lake. The concentrations of PAHs in the SPMD dialysates and fish tissues were analyzed by gas chromatography-mass spectrometry. In addition, the EROD activity of SPMD dialysates was assayed with H4IIE rat hepatoma cells and the induction of EROD activity in pancreas and liver of fish was also assayed. It was found that dialysates from SPMDs with longer exposures generally induced stronger activity and the toxic potency of dialysates expressed as bioassay-derived 2, 3, 7, 8-tetrachlorodibenzo-p-dioxin (TCDD) equivalents ranged from 3.8 pg/g to 6.2 pg/g for 32 days exposure. A correlation of toxicity equivalency quantity (TEQ) with chemical analysis and in vitro bioassay-derived TEQ(bio) of SPMD dialysates yielded an R2 of 0.88, therefore, indicated that PAHs were one of the important AhR agonists in the water. Empirical evidence suggests that PAHs can account for about 40% to 50% of the potency observed. Moreover, a good correlation was observed between the results of EROD bioassay in vivo and EROD bioassay in vitro (R2 = 0.62), thus reflected that these two methods can be complementarily each other. It is, therefore, suggested that the SPMD technique combined with chemical analyses and in vitro EROD assay might be a valuable monitoring tool to assess the levels and effects of AhR agonists in water ecosystem.

With root injection and foliar spray, this paper studied the effects of different concentrations salicylic acid, brassinolide, chitosan and spermidine on the growth, morphogenesis, and physiological and biochemical characters of cucumber ( Cucumis sativus L. ) seedlings under 200 mmol x L(-1) NaCl stress. The results showed that at proper concentrations, these four exogenous chemical activators could markedly decrease the salt stress index and mortality of cucumber seedlings, and the decrement induced by 0. 01 mg x L (-1) brassinolide was the largest, being 63. 0% and 75. 0% , respectively. The activities of superoxide dismutase (SOD) , peroxidase (POD) and catalase (CAT) increased significantly, resulting in a marked decrease of malondialdehyde (MDA) content and electrolyte leakage. The dry weight water content and morphogenesis of cucumber seedlings improved, and the stem diameter, leaf number, and healthy index increased significantly. All of these suggested that exogenous chemical activators at proper concentrations could induce the salt resistance of cucumber, and mitigate the damage degree of salt stress. The salt resistance effect of test exogenous chemical activators decreased in the sequence of 0.005 -0.05 mg (L-1) brassinolide, 150 -250 mg x L (-1) spermidine, 100 -200 mg x L(-1) chitosan, and 50 -150 mg x L(-1) salicylic acid.

940. [The influences of lactose as an inducer on expression of plant des-pGlu1-brazzein in Escherichia coli].
Expression strain of des-pGlu1-brazzein was constructed and the conditions using lactose as inducer was also optimized. The Influences of three factors which were lactose concentration, induction time and inducing temperature on the growth of strain and on the yield of des-pGlul-Brazzein was analyzed in detail. The result indicated that high lactose concentration inhibit the growth of strains (P < 0.01) but made no difference on expression of target protein between 0.5%-5% (P > 0.05), Biomass would be improved as time passed (P < 0.01), but the yield of target protein didn't increase obviously at 30 degrees C compared with at 37 degrees C. Further result showed that the greater expressed level of des-pGlul-Brazzein, as high as about 20% of total cell protein, could be achieved after the strain had been induced with 0.5% lactose under 28 degrees C - 30 degrees C for 4 h.
