3704. [Effects of differentiation status on apoptosis of human leukemia HL60 cells].

The effects of differentiation of human leukemia HL60 cells on harringtonine(Har) and camptothecin(Cam) induced apoptosis(in these cells) were studied. When treated with phorbol 12-myriate, 13-acetate 16 nmol.L-1 for 24 h, the HL60 cells differentiated into monocyte/macrophage cells and were arrested at G1 phase. The differentiated cells were shown to be resistant to the Har and Cam induced apoptosis, but showed no change of expression of c-myc gene. HL60 cells incubated in 1.4% dimethyl sulfoxide for 48 h differentiated into granulocyte cells and were also gene arrested at G1 phase. The differentiated cells became resistant to the apoptosis induced by Cam, but not that by Har, and expression of c-myc decreased drastically in the differentiated cells. The results indicate that the differentiated status of human leukemia HL60 cells apparently affected the apoptosis induced by harringtonine and camptothecin, but it was irrelevant to the change of the expression of c-myc gene.

3709. [Effects of lanthanum and cerium on malignant proliferation and expression of tumor-related gene].
Cultured human gastric cancer cell line PAMC82 was studied in vitro to further verify anti-tumor effect sof rare-earth elements and explore their mechanism of tumor inhibition. Inhibitory effects of elements lanthanum and cerium on cell growth, reverse effects of them on reduction of malignancy and effects of them on level of expression of oncogene and cancer suppressor gene were observed. Lanthanum chloride, cerium chloride and mixed rare-earth chloride at levels of 0.5 to 1.5 mmol/L could inhibit obviously growth of cancer cells and change cell morphology and microtubule structure of PAMC82, similar to that of normal cells, their colony-forming ability lowered in soft agar, and expression of tumor suppressor gene p53, p16 and p21 increased and that of gene nm23 lowered.
3710. [Inhibition of sodium selenite on transformation of human B lymphocytes by Epstein-Barr virus].
We detected the effect of non-toxic doses of sodium selenite (Se) on the expressing effect of nuclear antigen (EBNA) of Epstein-Barr virus (EBV) in Raji cell line by microfluorescence spectrophoto-quantitation (MFS) and studied the inhibing transformation in B lymphocytes from human umbilical cord blood (BL) with EBV in vitro by Se. We found that Se (0.01-0.50 microgram/ml) can lower EBNA-MFS value 8.4%-21.8% in Raji cell line. The transformation effect of BL with EBV was markedly inhibited by Se (0.1-1.0 microgram/ml) in pretreatment and cotreatment. The inhibition rates of BL transformation were 73.4%-92.1% and 60.3%-77.2% respectively. EBV is associated with nasopharyngeal carcinoma (NPC). The Se levels of serum and hair in NPC patients were markedly lower than those in healthy persons. The results suggest that supplement Se to persons for active expression of EBV gene and NPC patients may help protect againt and treat NPC and diseases associated with EBV.

3713. [Observation of relationship between gastric filling and vomiting during chemotherapy].
The purpose of studying the relationship between degree of gastric filling and vomiting during chemotherapy is to reduce the occurrence of vomiting. 42 patients were selected to give intravenous chemotherapy at 3-4 h after a small simple breakfast. The result showed that there was a relationship between the degree of gastric filling and vomiting during chemotherapy.

The method of mRNA differential display was employed to isolate expressing genes involved in taxol-induced apoptosis of human breast cancer cells (BCap 37). Results demonstrated that the expressive alteration of twelve clones isolated was associated with taxol-induced apoptosis identified by Northern blot. cDNA sequence of clone C3P3 was highly homologous (99%) to human S-adenosylmethionine (S-AdoMet) synthetase. Increased transcriptional level of clone C3P3 induced by taxol was parallel to enhanced enzyme activity of S-AdoMet synthetase. These results suggested that the expression of these clones isolated by the mRNA differential display be possibly involved in the regulation of taxol-induced apoptosis. Their function in details need further study.
3715. [Drug resistance mechanism of doxorubicin-resistant human gastric cancer cells BGC-823/DOX].
To further investigate drug resistance mechanism of BGC-823/DOX, we analysed the amplication, transcription and overexpression of genes associated with drug resistance by Southern blot, RT-PCR, immunohisto clogical assay and the concentration of Doxorubicin in the cells by flow cytometry. The results showed that parent cells of BGC-823 were cell line with intrinsic drug resistance similar to the gastrointestinal tumors. Multidrug resistance was the main mechanism of drug resistance in BGC-823/DOX. There was a significant difference between the doxorubicin-resistant cells line from parent cells with intrinsic drug resistance and from parent cells without intrinsic drug resistance in drug resistance.

Our previous work showed the existence of low molecular weight tumor suppressors in human fetal tissues. In this paper, two tumor cell suppressors were isolated and purified from methanol extract of human fetal liver by C18 reversed-phase medium pressure chromatography, gel filtration on Sephadex LH-20, and high-performance liquid chromatography, directed by suppression of growth of HL-60 cells. The structures of the suppressors were identified to be 7-ketocholesterol and 7-beta-hydroxycholesterol. Under the condition of in vitro agar plate culture, 7-ketocholesterol and 7-beta-hydroxycholesterol showed preferentially inhibitory effects on the growth of both human and murine leukemic cell lines including human HL-60 and murine S-180 cells, but less effective on the growth of normal human and murine bone marrow granulocyte-macrophage progenitors (CFU-GM).

In vitro, inhibition of 5 compounds from Bidens bipinnata on two leukemia cells HL-60 and V397 is presented by determination of MTT living cell in this paper. The result show that the compounds have the action of inhibition, IC50 on V937 < or = 60 micrograms/ml.
3719. [Reversal of multidrug resistance by cyproheptadine in KBV200 cells].
The cyproheptadine (CYP) reversal of multidrug resistance (MDR) and its mechanism in KBV200 cell line were studies. MTT assay showed that CYP 15.0 mumol.L-1 could reverse vincristine, adriamycin (ADR) and etoposide resistance in KBV200 cells by a factor of 5.5, 2.0 and 1.9, respectively. CYP appeared to have no influence on the cytotoxicity of 5-fluorouracil (5-FU) and melphalan (MEL) in the cells. These results indicate that CYP is a MDR reversing agent. CYP 15.0 mumol.L-1 increased the ADR accumulation in KBV200 cells from 0.68 +/- 0.03 microgram/10(6) cells to 1.36 +/- 0.08 micrograms/10(6) cells (P < 0.01). CYP 15.0 mumol.L-1 was shown to obviously increase rhodamine 123 (R123) accumulation in and decrease its efflux from the cells. There was no change in PGP dying intensity under immunocytochemical assay and in mdr1 RNA level through slot blot analysis in the KBV200 cells exposed continuously to CYP 15.0 mumol.L-1 for 72 h. These results suggest that CYP acts by inhibiting the pumping function of PGP.
3720. [A comparative study on the blood vessel sclerosing effect of pingyungmycin and Natrii morrhuates].
Twelve adult rabbits were used in this experiment. After posterion auricular veins were temporarily blocked, pingyungmycin and natrii morrhuatis were injected separately and their effect on the blood vessels was studied. The results showed that pingyungmycin caused endotheliac cell oedema, degeneration, vascular shrinkage, vascular wall thickening, while the effect of natrii morrhuatis on the blood vessels was thrombosis, organization of thrombus and closure of the lumen of the blood vessels. The sclerosing mechanisms of the two medicines and their clinical applications were discussed.
